INDUKSI KALUS PADI VARIETAS PAKETIH DENGAN PENAMBAHAN ZPT IAA DAN BAP SECARA IN VITRO

Rice (Oryza sativa L.) is a key commodity in Indonesia’s food security, highlighting the importance of tissue culture for local variety development. However, information on callus induction in the Paketih variety remains limited, despite its role in plant regeneration. This study evaluates optimal IAA and BAP concentrations for callus induction in Paketih rice.. The research was conducted at the Tissue Culture Laboratory of the Jember State Polytechnic from October to December 2025. This study used a factorial RAL design with 2 factors and 3 replicates, IAA at 0.1, 0.2, 0.3 mg/L and BAP at 1, 2, 3 mg/L. The results showed that BAP at 3 mg/l had a highly significant effect on callus weight (0.95 grams), while IAA at 0.1 mg/l had a significant effect on callus weight (0.87 grams). The interaction of IAA and BAP had no significant effect on callus diameter but resulted in compact, light brownish-white callus.
